Abstract

Bangkal stem bark (N. subdita) contains secondary metabolite compounds from polyphenols such as flavonoids, saponins, steroids, and tannins which have the potential as natural antioxidants. Antioxidants have a function to protect the body from the bad effects of free radicals that can cause skin damage. This study aimed to determine the IC50 value of methanolic extracts of Bangkal stem bark before being formulated and after being formulated into antioxidant lotions. In this study lotion was formulated with different concentration of stearic acid as emulgator : FI (2%), F2 (3%) & FII (5%). Determination of antioxidant activity was carried out using the DPPH method and ascorbic acid as a comparison. The result of IC50 value of methanolic extract of N. subdita stem bark is 34.1787 ± 0.2781 ppm, which is classified as a very active antioxidant. The result shows that the increase of stearic acid concentration affect the antioxidant activity, the higher concentration of stearic acid, the greater IC50 value. The results for lotions (FI,FII& FIII) IC50 value are 61.5593 ± 0.2673 ppm, 66.8051 ± 0.2317 ppm & 72.6253 ± 2.3117 ppm, respectively. All  formulas have IC50 range 50-100 ppm which are classified as active antioxidants. Based on the research it can be conclude that the methanolic extract of N. subdita stem bark has a very active antioxidant activity and the lotions classified as active antioxidant.
