Abstract


Haruan fish is one of wetland biodiversity resources which widely consumed by local people of South Kalimantan. Some information of these fish has not been reported including protein profile information. This study aims to identify the protein profile of the Haruan Fish (Channa striata) from South Kalimantan. Haruan fish collected from Nagara, Hulu Sungai Selatan and Gambut, Banjar District. The protein extracted from flesh with Tris EDTA Buffer and then precipitated with Ammonium sulphate. Protein concentration was measured by Bradford method and the protein isolate separated based on molecular weight using SDS-PAGE method. Based on the results, precipitation of fish protein is the most optimum in saturation of Ammonium sulphate 70-80% for fish from Nagara with protein content 5,34 mg/mL and 60-70% for fish from gambut with protein content 8,34 mg/mL. Based on the results of SDS-PAGE analysis, Haruan fish from Gambut have 27 protein band and from Nagara have 23 protein band. Where the molecular weight of protein band varied from 24-191 kDa. This results suggest the origin of geography or natural habitat affect to protein profiles and the data from protein profiles analysis can be provided for genetic diversity assement of Haruan fish.
