VIABILITAS SPERMATOZOA KERBAU KALIMANTAN SELATAN YANG DIPRESERVASI DENGAN PENGENCER NIRA AREN DAN BEBERAPA KONSENTRASI GLISEROL

Abstract


The objective of this study was to examine the effect of various glycerol concentrations in sugar palm juice extender on viability of Kalimantan Selatan buffalo spermatozoa preserved at 5oC.  Semen was collected by artificial vagina.  Fresh semen wasimmediately evaluated and divided into four tubes, then diluted with: 73% lactose-based extender + 20% egg yolk + 7% glycerol (LG-7) as control, 74% sugar palm juice + 20% egg yolk + 6% glycerol (SPJG-6), 73% sugar palm juice + 20% egg yolk + 7% glycerol (SPJG-7), and 72% sugar palm juice + 20% egg yolk + 8% glycerol (SPJG-8), respectively.  Diluted-semen was preserved in refrigerator at 5oC.  Spermatozoa viability including percentages of motile spermatozoa and live spermatozoa were evaluated every day for three days.  The results of this study showed that there were no differences between treatments on viability of Kalimantan Selatan buffalo spermatozoa until day-4 of preservation.  On the third day of preservation, percentages of motile and live spermatozoa were 46.25 and 58.25% for LG-7, 43.75 and 58.25% for SPJG-6, 45.00 and 52.50% for SPJG-7, 41.25 and 52.00% for SPJG-8.
